Production of Interspecific Somatic Hybrids between the Cultivated Potato (Solanum tuberosum) and the Wild Species (S. brevidens)





Abstract
A total of 322 somatic hybrid plants were obtained from protoplast fusion between Solanum brevidens and several clones of S. tuberosum (¢¥Superior¢¥, ¢¥Dejima¢¥, and dihaploid of ¢¥Superior¢¥). Putative somatic hybrids could be selected through the detection of polyploid by flow cytometry. Electro-fusion method was superior to PEG-mediated fusion methods in plating efficiency and production of somatic hybrid plants. In the electro-fusion, optimum CaCl©ü concentration in the fusion solution was 0.5 mM. In the PEG-mediated fusion experiment, plating efficiency and the frequency of the true somatic hybrid were increased by about 10% in the large scale fusion experiment as compared to the small scale fusion. True somatic hybrid plants could be selected through RAPD. Somatic hybrid plants could be identified through the observation of morphological characteristics. Somatic hybrid plants grew more vigorously and had broader leaves than cultivated potatoes.
